What would happen if the root cap of a growing root is removed?

The root cap protects the delicate meristematic region of the root from mechanical damage as it pushes through the soil. This follows from NCERT principle where relation explains outcome clearly for students.

Hypophysis cell contributes to:

Hypophysis represents uppermost cell of basal lineage derived from basal cell after first highly asymmetric zygotic division; it resides at junction between suspensor file and embryo proper. It undergoes crucial asymmetric division generating lens-shaped quiescent center precursor and columella stem cell precursor, thus directly founding root meristem. Specification requires auxin flux from embryo proper via basal PIN1 polarity, MP-ARF5 mediated transcriptional response, and high auxin-induced PLT accumulation and WOX5 induction. Mutants lacking auxin perception in hypophysis exhibit horizontal division and fail to establish functional root pole.

WOX5 contributes to:

WOX5 WUSCHEL-related homeobox 5 transcription factor is exclusively expressed in quiescent center cells of root apical meristem functioning analogously to shoot WUS. It moves non-cell-autonomously to surrounding cortex/endodermis and columella initials repressing differentiation-promoting transcription factor CYCLING DOF FACTOR 4 and maintaining stem cell identity. WOX5 sustains PLT gradient, auxin homeostasis, and represses differentiation via local chromatin modifications. Its expression maintained by SHR/SCR and PLT feedback as well as auxin, illustrating conserved WOX-based organizing center mechanism preserving root stem cell reservoir enabling continuous indeterminate growth.

Quiescent center characterized by:

Quiescent center of Arabidopsis root tip consists of four to seven cells characterized by extremely slow division rate, highly oxidized redox environment, condensed chromatin, and enriched expression of WOX5, QC25, QC46 markers, and elevated auxin signaling reporters. These rarely dividing organizer cells protect genomic integrity while preventing differentiation of surrounding initial cells through short-range mobile signals involving WOX5 and CLE40-CLV1 peptides. Upon severe wounding or stem cell ablation, QC cells rapidly re-enter cell cycle replenishing lost initials, revealing protective quiescence mechanism enabling reservoir function for regeneration and continuous root growth.

PLT mutants lack:

PLETHORA genes are master regulators specifying embryonic root fate downstream of auxin accumulation. Loss-of-function plt1 plt2 double and higher order plt mutants produce seedlings completely lacking embryonic root pole despite presence of auxin maximum and basal polarity. Quiescent center marker WOX5 never induced, columella stem cells differentiate prematurely, basal pole converts to hypocotyl-like tissue showing reduced PIN expression and ectopic shoot reporters. Since PLTs also promote PIN transcription, their absence destabilizes auxin transport further reinforcing rootless phenotype, demonstrating absolute requirement of auxin-induced PLT effectors.

Auxin-induced gene promoting root identity:

PLETHORA family AP2-domain transcription factors PLT1, PLT2, PLT3, PLT5, PLT7 are directly transcriptionally induced by auxin maxima at root pole through ARF5, ARF7, ARF10, ARF16 after auxin-triggered degradation of AUX/IAA repressors via TIR1/AFB pathway. PLT proteins accumulate in graded manner highest near quiescent center promoting stemness, intermediate sustaining mitotic activity, low permitting differentiation and elongation. PLTs activate WOX5, PIN genes, and cell cycle regulators while repressing shoot determinants. Auxin-PLT-PIN positive feedback stabilizes auxin maximum defining stem cell niche patterning and regeneration.

Root apical meristem contains:

Root apical meristem contains highly organized niche with quiescent center comprising rarely dividing organizer cells surrounded by distinct stem cell initials for epidermis, lateral root cap, columella, cortex/endodermis common initial, and stele. Immediate daughters proliferate rapidly in meristematic zone, transit through elongation zone where vacuolar expansion dramatically increases length, then differentiate in maturation zone forming root hairs, Casparian strips, and mature xylem. Thus meristem integrates self-renewing stem cells maintained by WOX5, PLT, SHR/SCR signals and differentiating progeny enabling continuous growth while preserving precise radial and longitudinal patterning.

SCR mutation results in:

In wild-type root meristem, cortex/endodermis initial stemming from stem cell niche undergoes formative asymmetric periclinal division producing distinct cortex and endodermis layers strictly controlled by SHR-SCR regulatory module. SCARECROW is essential to promote that division and to impose endodermal fate while repressing cortical program through downstream transcriptional activation. scr mutants fail to execute periclinal division, resulting single ground tissue layer surrounding stele. Molecular profiling reveals this solitary layer co-expresses cortical marker Co2 alongside endodermal markers CASP1 and ectopic lignin, indicating mixed identity and demonstrating necessity of SCR for both division and fate separation.

SHORT ROOT and SCARECROW regulate:

SHORT ROOT and SCARECROW are GRAS family transcription regulators establishing radial ground tissue pattern in Arabidopsis root meristem. SHR protein synthesized exclusively in stele moves via plasmodesmata into adjacent endodermal layer where it activates SCR transcription and forms SHR-SCR heterodimer that sequesters SHR preventing further movement. Complex triggers asymmetric periclinal division producing inner endodermis and outer cortex. Downstream IDD, MAGPIE, and SCL3 targets direct differentiation. Loss of either abolishes formative division, radial organization collapses, emphasizing mobile transcription factor mechanism converting positional signals into concentric tissue architecture surrounding vascular cylinder.

Rhizogenesis refers to formation of:

Rhizogenesis denotes formation of roots from callus, shoots or explants, derived from rhiza root, complementing caulogenesis in organogenesis. It is induced predominantly by elevated auxin levels and low cytokinin, mirroring natural lateral root formation. Molecular basis involves creation of localized auxin maxima through biosynthesis via YUCCA pathway and polar transport by PIN1, PIN2, AUX1 carriers concentrating auxin in founder cells. Perception via TIR1/AFB F-box receptors triggers degradation of Aux/IAA repressors liberating ARF7 and ARF19 transcription factors that induce LBD16, LBD29 and PLETHORA genes specifying pericycle-like cells to organize quiescent center expressing WOX5. Auxin also activates plasma membrane H+ ATPase causing apoplast acidification, expansin mediated wall loosening and initiation of primordia. Cytokinin antagonism is relieved when cytokinin level drops, allowing downregulation of SHY2 inhibitor and restoration of PIN expression. In tissue culture, IBA or NAA at 0.5-3 mg per L triggers adventitious root emergence within 10 days from shoot bases, essential for final rooting stage of micropropagated plantlets before hardening and field transfer ensuring autotrophic survival.
