NK cells employ missing-self recognition, detecting loss of inhibitory signals. Normal cells expressing self MHC class I engage KIRs and CD94/NKG2A inhibiting killing. Tumor or virus-infected cells downregulating MHC I escape T-cell recognition but become susceptible to NK cytotoxicity through reduced inhibition.
