O6-methylguanine-DNA methyltransferase removes alkyl adducts from O6 position of guanine by transferring methyl group to its own cysteine residue, irreversibly inactivating itself. Each enzyme molecule acts only once, hence designated suicide enzyme. This direct repair mechanism protects genome without excision and prevents mutagenic G to A transitions.
