A linear Scatchard plot is diagnostic of a single class of independent, non-cooperative binding sites. Under these conditions the simple one-site mass-action model holds, allowing unambiguous extraction of Kd from the slope and Bmax from the intercept. Curvature indicates either site heterogeneity or cooperative interactions and necessitates more complex analytical models.
