Prototrophs synthesize compounds de novo from simple precursors. They represent parental wild-type phenotype against which auxotrophic mutants blocked in specific pathways are scored. Prototroph selection on minimal plates underpins genetic mapping of mutations, recombination analysis, and complementation tests in bacteria and Neurospora.
