Mung bean nuclease is a single-strand-specific nuclease extracted from germinating sprouts of mung bean, Vigna radiata, a leguminous plant. Similar to S1 nuclease, it acts as an endonuclease preferentially degrading unpaired regions of DNA and RNA to mononucleotides, requiring zinc and acidic pH, while exhibiting limited activity on double-stranded substrates when used at low concentrations. The enzyme is valuable for trimming single-stranded extensions to generate blunt-ended cloning fragments, removing overhangs to improve ligation fidelity, and analyzing secondary structures of nucleic acids in vitro during genetic engineering workflows.
