Once inside, homologous sequences integrate via RecA-mediated recombination replacing recipient allele. Competence develops in Bacillus subtilis, Streptococcus pneumoniae, Haemophilus. Naked nature is proven by DNase sensitivity: addition of DNase destroys transforming activity, unlike phage-mediated transfer which remains DNase resistant and protected.
