Scatchard analysis converts hyperbolic saturation data into a linear form that simultaneously yields receptor affinity (from the slope) and total binding capacity (from the intercept). The method remains valuable provided equilibrium is attained, ligand depletion is negligible, and nonspecific binding is accurately subtracted. When these conditions are satisfied the plot provides a clear quantitative description of the ligand–receptor interaction.
