Signal amplification occurs when a single activated receptor molecule catalyzes the activation of many downstream molecules, each of which can in turn activate still more targets. Phosphorylation cascades and second-messenger systems are classic examples. Amplification enables a small number of ligand–receptor complexes to generate a large cellular response while preserving specificity through spatial and molecular constraints.
