Most molecular markers require prior sequence information for primer design, such as SSR, EST and SNP chip fabrication. Random Amplified Polymorphic DNA employs single short decamer primers of arbitrary sequence that anneal at low stringency to multiple genomic sites, generating multilocus profiles without any genome sequence knowledge. This attribute made RAPD valuable for orphan species and initial diversity surveys. However, dominance and reproducibility limitations restrict its use. Ability to amplify without prior sequence data is defining advantage for rapid preliminary polymorphism detection.
